What are some quests .sights of grace .or bosses that will lock me out of content or trigger world formation ?

If I burn the Erdtree will I still be able to fight Ranni and Maliketh

You have to fight maliketh to progress in the game. I'm not sure about ranni since I thought her quests were supposed to be done in my playthrough. Just play the game, don't worry about making sure you get whatever ending you read about on the internet on your first playthrough. That's what NG+ is for.
